GHK Basic is the tripeptide Gly-His-Lys in free form, without an attached copper ion. This is the original signaling molecule the body uses as an endogenous marker of tissue damage: when collagen breaks down, this fragment is released, triggering the fibroblast repair response. GHK levels in plasma are high in youth (approximately 200 ng/mL at ages 20–25) and decline to roughly 80 ng/mL by age 60, correlating with slower tissue recovery rates. The peptide regulates the expression of more than 1,500 genes, shifting their profile toward repair, reduced inflammation, and suppression of oxidative stress.

As a standalone molecule without copper, GHK retains the ability to activate TGF-β-dependent collagen synthesis pathways, interact with fibroblast receptors, and modulate matrix metalloproteinase activity — balancing breakdown and synthesis in the extracellular matrix. Nevertheless, copper is a key cofactor: the Cu²⁺ ion enhances the activity of lysyl oxidase and lysyl hydroxylase — enzymes that cross-link collagen and elastin — and without it, some effects are less fully realized. GHK Basic is rationally used where the copper ion is undesirable for formulation reasons or due to individual metal sensitivity.

Applications span skin structure support, accelerated soft tissue healing, and neuroprotection: in several studies GHK demonstrated anti-inflammatory and neurotrophic potential through regulation of genes linked to neuronal survival.
