HEP-1 (Human Ezrin Peptide 1) is a synthetic 14-amino-acid peptide reproducing amino acids 324–337 of the ezrin protein — a cytoskeletal protein linking the cell membrane to the actin cytoskeleton and participating in T-cell immune synapses. This region, called the Hep receptor, forms a double antiparallel alpha helix with alternating charged residues — a structure that plays a role in activation of the adaptive immune response. By binding to this ezrin region, HEP-1 modulates conformational changes in the protein and initiates downstream signaling events that enhance immune activity.
As an immunomodulator, HEP-1 enhances adaptive immunity through B- and T-cell arms: it increases antibody titers after hepatitis B vaccination, strengthens lymphocyte proliferative response, and normalizes subpopulation ratios in immunodeficient states.
