KP1 is a peptide derived from the Klotho protein โ one of the central regulators of biological aging. The Klotho protein was named after the goddess who spins the thread of life: its expression in the kidneys and brain predictably declines with age, and Klotho gene knockout mice demonstrate accelerated aging with osteoporosis, muscle atrophy, neurodegeneration, and shortened lifespan. KP1 was identified by screening a series of peptides from the Klotho structure and selected as the most biologically active fragment, reproducing the effects of the full-length protein in a compact format.
The central mechanism of KP1 at the cellular level is suppression of the TGF-ฮฒ1/Smad3/miR-223-3p signaling cascade, which under chronic activation (inflammation, ischemia, toxins) triggers senescence of renal tubular epithelial cells and fibrosis. KP1 restores endogenous Klotho expression at the post-transcriptional level: through blockade of miR-223-3p and lncRNA-TUG1, protein suppression is relieved, and the cell again produces its own Klotho in normal amounts. In chronic kidney disease (CKD) models, KP1 administration significantly reduced senescence markers (p21, p16, ฮณ-H2AX), decreased accumulation of SA-ฮฒ-Gal-positive senescent cells, and blocked fibrosis progression.
Beyond the kidneys, KP1 affects the systemic Klotho axis linked to neuroprotection, cardiovascular health, and suppression of chronic inflammation. Klotho via FGF23-dependent and independent pathways regulates phosphate and calcium metabolism, mitochondrial function in neurons, and endothelial NO production. KP1 as an active fragment reproduces part of this systemic signal. Human clinical data do not yet exist โ the molecule is at the stage of preclinical models and early translational research.
