Vesugen is a tripeptide Lys-Glu-Asp (KED), a vascular system bioregulator from the Khavinson series, developed at the St. Petersburg Institute of Bioregulation and Gerontology. Like other molecules in the series, Vesugen exerts its effect through direct DNA interaction: the peptide penetrates the cell nucleus and binds the minor groove of DNA in the CATC promoter region of the MKI67 gene encoding Ki-67 β a marker and regulator of cell proliferation. Reduced Ki-67 expression is characteristic of aging endothelial cells, leading to slowed vascular endothelium renewal. Through this epigenetic point, Vesugen restores proliferative potential of endothelial cells, renewing vessel lining at a rate comparable to a young organism.
An additional mechanism is SIRT1 (sirtuin 1) activation, an NADβΊ-dependent geroprotector: this directly links Vesugen to longevity programs, protection from oxidative stress, and limitation of atherosclerotic remodeling. In cellular models and in vivo, Vesugen normalized endothelin-1 expression (a vasoconstrictor elevated in atherosclerosis and restenosis), enhanced connexin-mediated coupling between endothelial cells, increased nitric oxide production, and restored vasodilatory reserve. In clinical observations in elderly patients with cardiovascular pathology, course use was associated with reduced angina frequency, fewer arrhythmias, and improved exercise tolerance.
A unique feature of Vesugen among Khavinson bioregulators is coverage of not only the vascular but also the neurological axis: via the SIRT1 pathway, the peptide regulates genes of neuronal differentiation and neuroprotection, and in studies on elderly individuals with CNS disorders, improvements in memory and cognitive performance were recorded. This makes Vesugen useful not only as a "vascular" bioregulator but also as a systemic anti-aging agent.
